Keap is a transcription factor that binds to the C-terminal domain of IκB proteins
Drip is a post-translational modification that alters the function of Keap, resulting in its dissociation from the IκB protein.
Processing of IκB occurs through two pathways, one mediated by the proteasome, the other by the peptidase. The proteasome-mediated pathway is responsible for the degradation of the N-terminal domain of certain members of the IκB subfamily, while the peptidase-mediated pathway is responsible for the degradation of the C-terminal domain. Drip acts by cleaving the peptidase-mediated IκB protein at a site within its N-terminal domain. This action results in the creation of a new site for Keap binding. In cells expressing both Keap and Drip, Keap binds to the newly created site on the C-terminal domain, resulting in the dissociation of Keap from IκB. As a result, IκB phosphorylation is reduced, and NF-κB translocates into the cell nucleus and initiates gene expression.
The integration of Keap and Drip allows for the maintenance of optimal levels of NF-κB activation. In addition, it can be used to mediate temporary changes in NF-κB activation levels. In some cell types, such as endothelial cells, normal IκB function may be required to prevent excessive NF-κB activation. In these cells, Keap binding to IκB will result in the desensitization or inhibition of NF-κB. When cells are exposed to cytokines, which induce Drip expression, Drip will cleave IκB proteins, resulting in a reduction in NF-κB activity. After a period of time when cytokine levels have returned to normal levels, Keap is able to re-activate NF-κB via binding to newly created sites on the C-termini of IκB proteins.
