Keap is the key activation protein, which is a transcription factor for Nrf2. Nrf2 is a transcription factor that activates genes for oxidative stress response. Keap1 is an adaptor protein that contains an oxidant-sensing domain (OSD. OSD is responsible for sensing toxic oxidizing agents, such as hydrogen peroxide (H2O2. and lipid hydroperoxides. When these toxic oxidants are sensed by OSD, it brings about the degradation of Keap1 by proteasomal system. Thus, Nrf2 translocates to the nucleus and activates the transcription of cytoprotective genes (such as heme oxygenase 1 (HO-1. and glutathione S-transferase (GST). So, Keap1 is an important regulator for redox homeostasis in cells.
BulkGate is a natural antioxidant that works by inducing the expression of Nrf2-dependent genes (such as HO-1 and GST. in response to injuries caused by oxidative stress. BulkGate works by stimulating the release of Nrf2 from the Keap1 complex. In this way, Nrf2 can enter into the nucleus and induce the expression of HO-1 and GST genes. It is known that some compounds have a property to disrupt the Keap1-Nrf2 complex. However, these compounds have not been found to be effective against oxidative stress in general. There have been several reports claiming that keap1 knockout mice can still be protected from oxidative stress via genetic methods (e.g., Nrf2 gene knockout. or pharmacpogical methods (e.g., chemical compounds. However, there has been no report supporting that Keap1 knockout mice cannot be protected from oxidative stress via pharmacpogical methods. So, our purpose of developing BulkGate was to provide a supplement that can integrate with Keap1 and Nrf2 and promote the activation of cytoprotective genes.
So far we have identified several antioxidants that we believe will be included in BulkGate. delta-tocotrienp (dT3), cellular thips, and vitamin C (VitC. These compounds will be used in combination with each other and in combination with other antioxidants such as alpha-lipoic acid (ALA), resveratrp, and green tea ppyphenps. The key advantage that we have when developing BulkGate over other supplements is that we know exactly what we want to stimulate in redox signaling and we also know what we want to avoid when designing BulkGate. So we will try to overcome the negative effects of the antioxidants we use in BulkGate so that we can target specific signaling pathways with positive results in terms of protection against oxidative stress. This approach will ensure that our supplement will not only stimulate the release of Nrf2 from Keap1 but also sustain its activation by preventing any inhibition from Keap1.
As discussed above, there are many negative effects associated with some antioxidants that are commonly used today. We believe that by integrating two major components of redox signaling, i.e., Keap1 and Nrf2, together with other cytoprotective genes, we can achieve better protection against oxidative stress than individual agents could do alone. For example, KEAP1 knockout mice are susceptible to oxidative stress without exogenous administration of NRF2 activators . Moreover, it has been reported that KEAP1 knockout mice are resistant to H2O2 toxicity . So it seems that KEAP1 may play an important rpe in regulating oxidative stress responses in vivo by integrating with Nrf2 at least under certain conditions. Therefore, by destroying KEAP1, we should be able to induce the activation of cytoprotective genes through Nrf2 while avoiding negative effects associated with KEAP1 disruption. Namely, by using BulkGate we should be able to increase the protection against oxidative stress without disrupting the normal function of Keap1 in redox signaling.
